Product Description
Cagrilintide is a long-acting synthetic amylin analog, a 37-amino-acid peptide whose backbone is derived from the human amylin analog pramlintide (itself based on human amylin / islet amyloid polypeptide). Its sequence incorporates several amino-acid substitutions and an intramolecular disulfide bridge between two cysteine residues, together with a lipidation (fatty-acid acylation) modification that promotes reversible binding to serum albumin. This acylation chemistry is the principal feature that distinguishes it from native amylin, markedly extending its circulating half-life in research models and giving rise to its characterization as a long-acting amylin analog.
In preclinical and in-vitro studies, Cagrilintide has been investigated as a non-selective amylin receptor agonist, with reported activity at the calcitonin receptor and the RAMP-containing amylin receptor complexes (the AMY1, AMY2, and AMY3 subtypes). It has been used as a tool compound to study satiety signaling, energy homeostasis, and body-weight regulation, with animal-model research pointing to hindbrain regions such as the area postrema and nucleus of the solitary tract as relevant sites of activity. Investigations have also examined its co-administration with incretin-class peptides in research settings exploring metabolic pathways.
