Hexarelin

Hexarelin is a synthetic growth hormone-releasing peptide (GHRP) belonging to the class of growth hormone secretagogues. It is a hexapeptide composed of six amino acids with the sequence His-D-2-methyl-Trp-Ala-Trp-D-Phe-Lys-NH2, and is structurally derived from GHRP-6, incorporating a 2-methyl-D-tryptophan substitution that distinguishes it from earlier secretagogue analogs. As a laboratory reference compound, it is studied as a synthetic ligand at the growth hormone secretagogue receptor (GHS-R1a), the same receptor targeted by the endogenous hormone ghrelin.

In preclinical and in-vitro studies, Hexarelin has been investigated for its role in stimulating pulsatile growth hormone release from anterior pituitary tissue, where animal models have characterized it as one of the more potent secretagogues among the GHRP family. Beyond its pituitary activity, research has explored interactions with the CD36 scavenger receptor expressed in cardiac tissue. In rodent ischemia-reperfusion models, investigators have reported observations relating to cardiomyocyte survival, oxidative stress markers, and inflammatory signaling pathways, with some studies noting effects that appeared independent of the growth hormone axis.
