IGF-1 LR3 (Long Arg3 Insulin-like Growth Factor-1) is a synthetic, single-chain polypeptide analogue of human insulin-like growth factor 1 (IGF-1). It is engineered with two structural modifications relative to the 70-residue native protein: a substitution of arginine for glutamic acid at the third position and a 13-amino-acid extension at the N-terminus, giving a total length of 83 amino acids. These changes markedly reduce its affinity for the insulin-like growth factor-binding proteins (IGFBPs) while retaining agonist activity at the IGF-1 receptor, characteristics that make it a useful reference ligand in growth-factor biology.

In preclinical and in-vitro research, IGF-1 LR3 has been investigated as an agonist of the IGF-1 receptor (IGF-1R) and as a supplement in serum-free cell-culture systems. Reported laboratory observations include activation of downstream PI3K/Akt and MAPK/ERK signaling cascades and associated effects on cell proliferation, protein-synthesis pathways, and anti-apoptotic signaling in cultured cells. Because of its low IGFBP binding, researchers have noted increased availability of free ligand to target-cell receptors in these model systems compared with native IGF-1.

In animal and tissue-model studies, IGF-1 LR3 has been examined for its role in myoblast and satellite-cell proliferation, myotube differentiation, and tissue-repair pathways.
