Ipamorelin

Ipamorelin is a synthetic pentapeptide growth hormone secretagogue, composed of five amino acid residues with the sequence Aib-His-D-2-Nal-D-Phe-Lys-NH2 (alpha-aminoisobutyric acid, histidine, D-2-naphthylalanine, D-phenylalanine, and a C-terminally amidated lysine). Originally developed during research into ghrelin-mimetic compounds, it belongs to the growth hormone-releasing peptide (GHRP) family and acts as a selective agonist of the growth hormone secretagogue receptor type 1a (GHS-R1a), also known as the ghrelin receptor.

In preclinical and in-vitro studies, Ipamorelin has been investigated for its ability to stimulate pulsatile growth hormone release from somatotroph cells of the anterior pituitary via GHS-R1a activation and intracellular calcium mobilization. Animal-model research has characterized it as one of the more selective secretagogues in its class, reporting growth hormone release without significant elevation of cortisol, ACTH, or prolactin even at doses well above the GH-releasing threshold observed in those models. This selectivity profile has made it a frequently referenced reference compound in receptor-binding and secretagogue-selectivity studies.

Additional preclinical literature has examined Ipamorelin in the context of skeletal and metabolic research. In rodent studies it has been associated with increases in bone mineral content and longitudinal bone growth, and it has been investigated for its capacity to counteract glucocorticoid-induced reductions in bone formation in adult rat models. These observations are derived strictly from animal and laboratory research and have not been established in humans.
