Product Description
Retatrutide (research designation LY3437943) is a synthetic, lipidated single-chain polypeptide and a member of the incretin-mimetic peptide class. Structurally it is a 39-amino-acid peptide built on a glucose-dependent insulinotropic polypeptide (GIP) backbone scaffold and engineered with several non-coded residues (including alpha-aminoisobutyric acid substitutions) together with a C20 fatty diacid moiety that supports albumin binding and enhanced in-solution stability. It was originally developed by Eli Lilly as a research molecule and is widely characterized in the literature as a unitary triple receptor agonist with affinity for the glucagon-like peptide-1 receptor (GLP-1R), the GIP receptor (GIPR), and the glucagon receptor (GCGR).
In preclinical and in-vitro receptor pharmacology studies, retatrutide has been investigated for its simultaneous engagement of these three incretin and glucagon receptor pathways, and structural biology work has examined how a single peptide can manifest balanced agonism across all three targets. In animal models of diet-induced obesity, it has been studied for its observed effects on body-weight trajectories and energy expenditure, with researchers attributing part of the energy-expenditure signal to glucagon-receptor activation. Comparative rodent studies have also been used to contrast its activity profile against other multi-receptor incretin peptides.
Additional preclinical and translational research has explored retatrutide in the context of metabolic and hepatic models, including studies of glucose handling and markers associated with steatotic liver phenotypes. All such findings are reported strictly as experimental observations in laboratory, cell-based, and animal systems and are presented here for research characterization only.
