SS-31

SS-31, also known as elamipretide (MTP-131, Bendavia), is a synthetic aromatic-cationic tetrapeptide belonging to the Szeto-Schiller (SS) family of mitochondria-targeted peptides. Its structure comprises four amino acids in the sequence D-Arg-Dmt-Lys-Phe-NH2, where Dmt denotes 2',6'-dimethyltyrosine. The alternating aromatic and basic residues, together with the C-terminal amide and the non-natural D-arginine, give the molecule a net positive charge while reducing susceptibility to enzymatic degradation. This water-soluble peptide is reported in the research literature to penetrate cell and organelle membranes and to concentrate selectively within the inner mitochondrial membrane.

In preclinical and in-vitro studies, SS-31 has been investigated for its affinity to cardiolipin, a signature phospholipid of the inner mitochondrial membrane. Researchers have examined how this interaction may influence cytochrome c electron-transport behavior, mitochondrial membrane integrity, and the generation of reactive oxygen species in cultured cells and isolated mitochondria. In animal and cellular models, the peptide has been studied in the context of mitochondrial bioenergetics, ATP synthesis, and oxidative-stress pathways relevant to cardiac, renal, and neurodegeneration research. All such findings derive from laboratory and animal experiments and have not been established as human health outcomes.
